    Variation on the Kolmogorov Forcing: Asymptotic Dissipation Rate Driven by Harmonic Forcing

    The relation between the shape of the force driving a turbulent flow and the upper bound on the dimensionless dissipation factor β\beta is presented. We are interested in non-trivial (more than two wave numbers) forcing functions in a three dimensional domain periodic in all directions. A comparative analysis between results given by the optimization problem and the results of Direct Numerical Simulations is performed. We report that the bound on the dissipation factor in the case of infinite Reynolds numbers have the same qualitative behavior as for the dissipation factor at finite Reynolds number. As predicted by the analysis, the dissipation factor depends strongly on the force shape. However, the optimization problem does not predict accurately the quantitative behavior. We complete our study by analyzing the mean flow profile in relation to the Stokes flow profile and the optimal multiplier profile shape for different force-shapes. We observe that in our 3D-periodic domain, the mean velocity profile and the Stokes flow profile reproduce all the characteristic features of the force-shape. The optimal multiplier proves to be linked to the intensity of the wave numbers of the forcing function.Comment: 11 pages, 4 Figure

    Small scale dynamics of a shearless turbulent/non-turbulent interface in dilute polymer solutions

    We study the physics of the turbulent/non-turbulent interface (TNTI) of an isolated turbulent region in dilute polymer solutions and Newtonian fluids. We designed an experimental setup of a turbulent patch growing in water/dilute polymer solutions, without mean shear and far from the walls. The observations from the experiments are complemented and expanded by simulations performed using a localised homogeneous forcing to generate the turbulent front and the Finitely Extensible Elastic model with the Peterlin closure model for the polymer stress. The comparison, which shows that when Newtonian and viscoelastic TNTIs are fed by the same energy they behave in similar manner both in the experiments and in the simulations, permits to extend the applicability, on a qualitative basis, of single relaxation time polymer models also to turbulent/non-turbulent interfaces. From the detailed analysis offered by the numerical results, the alterations in the dynamics between strain and vorticity help understanding the mechanics of the polymer action on the TNTI without mean shear. The reduced vorticity stretching and increased vorticity compression terms are found to be due to the modified degrees of alignment between vorticity, polymer conformation tensor, and rate-of-strain tensor eigenvectors observed especially near the interface. These alignments at the smallest scales of the non-Newtonian turbulent flow lead to a reduced production of enstrophy and consequently to a reduced entrainment, which in this problem are seen as reduced advancement of a turbulent region

    Circulating adrenomedullin estimates survival and reversibility of organ failure in sepsis: the prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock-1 (AdrenOSS-1) study

    Background: Adrenomedullin (ADM) regulates vascular tone and endothelial permeability during sepsis. Levels of circulating biologically active ADM (bio-ADM) show an inverse relationship with blood pressure and a direct relationship with vasopressor requirement. In the present prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock 1 (, AdrenOSS-1) study, we assessed relationships between circulating bio-ADM during the initial intensive care unit (ICU) stay and short-term outcome in order to eventually design a biomarker-guided randomized controlled trial. Methods: AdrenOSS-1 was a prospective observational multinational study. The primary outcome was 28-day mortality. Secondary outcomes included organ failure as defined by Sequential Organ Failure Assessment (SOFA) score, organ support with focus on vasopressor/inotropic use, and need for renal replacement therapy. AdrenOSS-1 included 583 patients admitted to the ICU with sepsis or septic shock. Results: Circulating bio-ADM levels were measured upon admission and at day 2. Median bio-ADM concentration upon admission was 80.5 pg/ml [IQR 41.5-148.1 pg/ml]. Initial SOFA score was 7 [IQR 5-10], and 28-day mortality was 22%. We found marked associations between bio-ADM upon admission and 28-day mortality (unadjusted standardized HR 2.3 [CI 1.9-2.9]; adjusted HR 1.6 [CI 1.1-2.5]) and between bio-ADM levels and SOFA score (p < 0.0001). Need of vasopressor/inotrope, renal replacement therapy, and positive fluid balance were more prevalent in patients with a bio-ADM > 70 pg/ml upon admission than in those with bio-ADM ≤ 70 pg/ml. In patients with bio-ADM > 70 pg/ml upon admission, decrease in bio-ADM below 70 pg/ml at day 2 was associated with recovery of organ function at day 7 and better 28-day outcome (9.5% mortality). By contrast, persistently elevated bio-ADM at day 2 was associated with prolonged organ dysfunction and high 28-day mortality (38.1% mortality, HR 4.9, 95% CI 2.5-9.8). Conclusions: AdrenOSS-1 shows that early levels and rapid changes in bio-ADM estimate short-term outcome in sepsis and septic shock. These data are the backbone of the design of the biomarker-guided AdrenOSS-2 trial. Trial registration: ClinicalTrials.gov, NCT02393781. Registered on March 19, 2015

    45S rDNA Regions Are Chromosome Fragile Sites Expressed as Gaps In Vitro on Metaphase Chromosomes of Root-Tip Meristematic Cells in Lolium spp

    BACKGROUND: In humans, chromosome fragile sites are regions that are especially prone to forming non-staining gaps, constrictions or breaks in one or both of the chromatids on metaphase chromosomes either spontaneously or following partial inhibition of DNA synthesis and have been well identified. So far, no plant chromosome fragile sites similar to those in human chromosomes have been reported. METHODS AND RESULTS: During the course of cytological mapping of rDNA on ryegrass chromosomes, we found that the number of chromosomes plus chromosome fragments was often more than the expected 14 in most cells for Lolium perenne L. cv. Player by close cytological examination using a routine chromosome preparation procedure. Further fluorescent in situ hybridization (FISH) using 45S rDNA as a probe indicated that the root-tip cells having more than a 14-chromosome plus chromosome fragment count were a result of chromosome breakage or gap formation in vitro (referred to as chromosome lesions) at 45S rDNA sites, and 86% of the cells exhibited chromosome breaks or gaps and all occurred at the sites of 45S rDNA in Lolium perenne L. cv. Player, as well as in L. multiflorum Lam. cv. Top One. Chromatin depletion or decondensation occurred at various locations within the 45S rDNA regions, suggesting heterogeneity of lesions of 45S rDNA sites with respect to their position within the rDNA region. CONCLUSIONS: The chromosome lesions observed in this study are very similar cytologically to that of fragile sites observed in human chromosomes, and thus we conclude that the high frequency of chromosome lesions in vitro in Lolium species is the result of the expression of 45S rDNA fragile sites. Possible causes for the spontaneous expression of fragile sites and their potential biological significance are discussed
